The Canine Interleukin 6 (Dog IL-6) ELISA quantitates Dog IL-6 in canine serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Dog IL-6. Principle of the method The Canine IL-6 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Interleukin-6 (IL-6) ist ein multifunktionales 26 kD-Protein, das ursprünglich in Medium mit RNA-stimulierten Fibroblastenzellen entdeckt wurde. IL-6 scheint direkt an den Reaktionen beteiligt zu sein, die nach einer Infektion oder Zellverletzung auftreten, und kann sich als genauso wichtig wie IL-1 und TNF-alpha für die Regulierung der Akutphasen-Reaktion erweisen. IL-6 wird Beobachtungen zufolge von Fibroblasten, aktivierten T-Zellen, aktivierten Monozyten oder Makrophagen und Endothelzellen erzeugt. IL-6 beeinflusst eine Vielzahl von Zellen, darunter myeloische Vorläuferzellen, T-Zellen, B-Zellen und Hepatozyten. Außerdem scheint IL-6 bei der Proliferation von T-Lymphozyten mit IL-2 zu interagieren. IL-6 potenziert die proliferative Wirkung von IL-3 auf multipotenzielle hämatopoetische Vorläuferzellen. IL-6 spielt eine entscheidende Rolle bei der B-Zell-Differenzierung zu Plasmazellen und ist ein potenter Wachstumsfaktor für Plasmozytom und Myelom. IL-6 ist eine sehr nützliche Kulturergänzung zur Erzeugung einer hohen Anzahl von Antikörper-produzierenden Hybridomen. IL-6 wird hauptsächlich an Stellen mit akuter und chronischer Entzündung produziert und in das Serum abgesondert. Es induziert eine transkriptionelle Entzündungsreaktion durch den Interleukin-6-Rezeptor-alpha.
