Beschreibung
The Equine Interleukin-10 (Eq IL-10) ELISA quantitates Eq IL-10 in equine serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Eq IL-10. Principle of the method The Equine IL-10 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Interleukin-10 ist ein pleiotropes Zytokin, das eine wichtige Rolle als Regulator für die Funktion der lymphoiden und myeloischen Zellen spielt. Beim Menschen wird IL-10 durch aktivierte CD8(+) T-Zellen im peripheren Blut durch Klone von CD4(+)-T-Helferzellen erzeugt. Aufgrund der Fähigkeit von IL-10, die Zytokinsynthese und mehrere Funktionen der akzessorischen Zellen von Makrophagen zu blockieren, ist dieses Zytokin ein potenter Suppressor der Effektorfunktionen von Makrophagen, T-Zellen und NK-Zellen. Darüber ist IL-10 an der Regulierung der Proliferation und Differenzierung von B-Zellen, Mastzellen und Thymozyten beteiligt. Die Primärstruktur von humanem IL-10 wurde durch Klonierung der cDNA bestimmt, die für das Zytokin codiert. Das entsprechende Protein besteht aus 160 Aminosäuren mit einem vorhergesagten Molekulargewicht von 18.5 kDa. Aufgrund seiner Primärstruktur ist IL-10 ein Mitglied der Vier-α-Helix-Bündel-Zytokinfamilie. In Lösung ist humanes IL-10 ein Homodimer mit einem scheinbaren Molekulargewicht von 39 kDa. Obwohl es eine N-Glykosylierungsstelle enthält, fehlen ihm nachweisbare Kohlenhydrate. Rekombinantes Protein, das in E. coli exprimiert wird, behält somit alle bekannten biologischen Aktivitäten bei. Das menschliche IL-10-Gen befindet sich auf Chromosom 1 und liegt als Einzelkopie im Genom vor. Humanes IL-10 zeigt eine starke DNA- und Aminosäure-Sequenzhomologie für das Maus-IL-10 und einen offenen Leserahmen (ORF) im Genom des Epstein-Barr-Virus, BCRF1, das viele der biologischen Aktivitäten von zellulären Zytokinen aufweist und daher möglicherweise eine Rolle bei der Virus-Wirt-Interaktion spielt. Die immunsuppressiven Eigenschaften von IL-10 deuten auf einen möglichen klinischen Nutzen von IL-10 bei der Unterdrückung von Transplantatabstoßungen hin.
