The Human HSPA4 (HSP70) ELISA quantitates Hu HSPA4 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu HSPA4. Principle of the method The Human HSPA4 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Die HSP 70-Gruppe ist aus vier hochgradig konservierten Proteinen zusammengesetzt: HSP 70, HSC 70, GRP 75 und GRP 78. Eine starke Induktion der HSP 70-Expression erfolgt als Reaktion auf Hitzestress. HSP 70 und HSC 70 spielen eine Schlüsselrolle im Importsystem des zytosolischen endoplasmatischen Retikulums und der Mitochondrien. Sie sind sowohl im Zytosol als auch im Zellkern von Säugetierzellen vorhanden. GRP 78 befindet sich im endoplasmatischen Retikulum, wo es importierte sezernierte Proteine erhält und an der Faltung und Translokation von naszenten Peptidketten beteiligt ist. Die GRP 75-Expression beschränkt sich auf die mitochondriale Matrix und unterstützt die Translokation und Faltung von naszenten Polypeptidketten sowohl nukleärer als auch mitochondrialer Herkunft. Es wurde postuliert, dass Mitglieder der HSP 70-Familie als kraftgenerierende Motoren fungieren und sich für ihre Aktivität auf die Hydrolyse von ATP verlassen.
