The Human Activin A ELISA quantitates Hu Activin A in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Activin A. Principle of the method The Human Activin A solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Activine sind dimerische Wachstums- und Differenzierungsfaktoren, die zur Transformationsfamilie der strukturell verwandten Signalproteine des beta-Wachstumsfaktors (TGF-beta) gehören. Activin signalisiert durch einen heteromeren Komplex von Rezeptor-Serinkinasen, der mindestens zwei Typ I- (I und IB) und zwei Typ II (II und IIB)-Rezeptoren enthält. Alle diese Rezeptoren sind Transmembranproteine, die aus einer liganbindenden extrazellulären Domäne mit Zystein-reicher Region, einer Transmembrandomäne und einer zytoplasmatischen Domäne mit vorhergesagter Serin-/Threonin-Spezifität bestehen. Typ I-Rezeptoren sind für die Signalisierung von Bedeutung; Typ II-Rezeptoren sind für die Bindung von Liganden und für die Expression von Typ I-Rezeptoren erforderlich. Typ I- und II-Rezeptoren bilden nach der Liganbindung einen stabilen Komplex, der durch Typ II-Rezeptoren zur Phosphorylierung von Typ I-Rezeptoren führt.
