The Human Bone Morphogenic Protein 2 (Hu BMP-2) ELISA quantitates Hu BMP-2 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu BMP-2. Principle of the method The Human BMP-2 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Knochenmorphogene Proteine (BMP) sind Mitglieder der TGF-beta-Superfamilie, die die Knochen- und Knorpelbildung beeinflussen (Hogan 1996, Reddi 1998 und Francis-West et al. 1999). Reife BMPs sind 30 bis 38 kDa große Proteine, die eine TGF-beta-ähnliche Cystein-Knotenkonfiguration annehmen. Lovostatin verbessert die Knochenbildung durch Aktivierung des bmp-2Gens (Mundy et al. 1999). BMPs stimulieren die Produktion spezifischer Knochenmatrixproteine und verändern die Stromazellen- und Osteoklastenproliferation (Macias et al. 1999, Lecanda et al. 1997). BMPs können auch ein wichtiger Faktor für die Entwicklung der inneren Organe sein, mit Funktionen bei Zellvermehrung, Apoptose, Differenzierung und Morphogenese (Hogan 1996, Dale und Wardle 1999). BMPs scheinen für die normale dorsale/ventrale Strukturierung verantwortlich zu sein. Wie TGF-beta binden BMPs an einen Typ-II-Rezeptor, der dann die transduzierende Typ-I-Rezeptoreinheit rekrutiert und den Smad-Protein-Signalweg aktiviert (Massague 1994, Derynck 1997, Attisano 1993).Spezifikation
