The Human Monocyte Chemoattractant Protein-4/CCL1 (Hu MCP-4) ELISA quantitates Hu MCP-4 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu MCP-4. Principle of the method The Human MCP-4 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Zytokine sind eine Familie von sezernierten Proteinen, die an immunoregulatorischen und entzündlichen Prozessen beteiligt sind. Das Gen, das MCP-4/CCL13 codiert, ist eines von mehreren Cys-Cys (CC)-Zytokin-Genen, die in Clustern auf dem q-Arm von Chromosom 17 auftreten. Die CC-Zytokine sind Proteine, die durch zwei benachbarte Zysteine charakterisiert sind. Das von diesem Gen codierte Zytokin zeigt chemotaktische Aktivität gegenüber Monozyten, Lymphozyten, Basophilen und Eosinophilen, jedoch nicht gegenüber Neutrophilen. MCP-4/CCL13 signalisiert über CCR2B- und CCR3-Rezeptoren und spielt bei der Anreicherung von Leukozyten an der Stelle von allergischen und nicht-allergischen Entzündungen eine Rolle. Es ist auch an der Rekrutierung von Monozyten in der Arterienwand während des Krankheitsverlaufs von Atherosklerose beteiligt und spielt möglicherweise auch bei der Anziehung von Monozyten in Geweben, die chronisch exogenen Pathogenen ausgesetzt sind, eine Rolle.Spezifikation
