The Human Cystatin C (CST3) ELISA quantitates Hu Cystatin C in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Cystatin C. Principle of the method The Human Cystatin C solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Cystatin C ist ein Zystein (Thiol)-Protease-Inhibitor, der zur Typ II-Cystatin-Gen-Superfamilie gehört und der am häufigsten vorkommende extrazelluläre Inhibitor von Zystein-Proteasen ist. Cystatin C ist ein konstitutiv sezerniertes, amyloidogenes Protein, das einen zweifach symmetrischen Dimer bildet und sowohl die Aktivität der Zystein-Protease als auch die Expression von Klasse-II-MHC-Molekülen moduliert. Die Expression von Cystatin C ist ein Indikator für die Nierenfunktion und die glomeruläre Filtrationsrate. Mutationen im Cystatin-C-Gen können zu Proteinaggregaten führen, die an der hereditären Amyloid-Angiopathie (HCCAA) und Gehirnblutungen beteiligt sind. Obwohl sowohl wildes als auch mutiertes Cystatin C in der Lage sind, konzentrationsabhängige inaktive Dimere zu bilden, dimerisiert das mutierte Cystatin C in niedrigeren Konzentrationen und ist anfälliger für Serinproteasen, was die Aggregation erleichtern kann. In neuronalen Zellen stimuliert oxidativer Stress die Expression von Cystatin C, was die Apoptose positiv regulieren kann.
