The Human DMP1 ELISA quantitates Hu DMP1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu DMP1. Principle of the method The Human DMP1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. DMP-1 (Dentin Matrix Protein-1), auch als Dentin Matrix Acidic Phosphoprotein-1 bekannt, ist ein Familienmitglied der kleinen Integrin-Ligand-, N-gebundenen Glykoprotein-Familie. Es ist wichtig für die Mineralisierung von Knochen und Dentin. DMP-1 wird in Knochen, Zähnen und hypertrophischem Knorpel exprimiert. Es wird durch Präosteoblasten synthetisiert und enthält viele saure Domänen. DMP-1 lokalisiert den Zellkern undifferenzierter Osteoblasten, wo es als Transkriptionsregulator für die Osteoblasten-spezifische Genaktivierung fungiert und die Osteoblastdifferenzierung induziert. Während der Osteoblastenreifung erfährt DMP-1 eine Konformationsänderung und wird als Reaktion auf einen Zustrom von Calciumionen in den Zellkern durch Kaseinkinase II phosphoryliert. DMP-1 wird dann in die extrazelluläre Matrix (EZM) exportiert, wo es die Nukleation von Hydroxylapatit und die Bildung von kalzifiziertem Gewebe reguliert. DMP-1 wird proteolytisch zu N- und C-terminalen Fragmenten in der EZM von Knochen und Dentin verarbeitet. Das Protein wurde außerdem im Knochen als Proteoglykan mit hohem Molekulargewicht identifiziert, das aus dem N-terminalen DMP-1-Fragment und Chondroitinsulfat besteht. Der Verlust von DMP-1 kann zu hypomineralisierten Knochen führen.Spezifikation
