The Human Fibroblast Growth Factor 19 (Hu FGF19) ELISA quantitates Hu FGF19 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu FGF19. Principle of the method The Human FGF19 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Der Fibroblasten-Wachstumsfaktor-1 (FGF-1), auch als saurer FGF bekannt, und der Fibroblasten-Wachstumsfaktor-2 (FGF-2), auch als basischer FGF bekannt, sind Mitglieder einer Familie von Wachstumsfaktoren, die die Proliferation von Zellen mesenchymalen, epithelialen und neuroektodermalen Ursprungs stimulieren. Weitere Mitglieder der FGF-Familie sind die Onkogene FGF-3 (Int2) und FGF-4 (hst/Kaposi), FGF-5, FGF-6, FGF-7 (KGF), FGF-8 (AIGF), FGF-9 (GAF) und FGF-10-FGF-23. Die Mitglieder der FGF-Familie weisen eine zu 30–55 % identische Aminosäuresequenz und eine ähnliche Genstruktur auf. Sie sind in der Lage, kultivierte Zellen zu transformieren, wenn sie in transfizierten Zellen überexprimiert werden. Die zellulären Rezeptoren für FGFs sind Mitglieder einer zweiten Multigenfamilie, darunter vier Tyrosinkinasen, die als Flg (FGFR-1), Bek (FGFR-L), TKF und FGFR-3 bezeichnet werden.
