The Human Furin ELISA quantitates Hu Furin in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Furin. Principle of the method The Human Furin solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Furin ist eine membranassoziierte, calciumabhängige Serinprotease, die zur Subtilisin-ähnlichen Prohormonkonvertase (PC)-Familie gehört. Mitglieder dieser Zellenzymfamilie spalten die meisten Prohormone und Neuropeptidvorstufen auf. Zahlreiche weitere zelluläre Proteine, einige virale Proteine und bakterielle Toxine, die über den konstitutiven Sekretionsweg transportiert werden, sind ebenfalls ein Reifungsziel für PCs. Furin und andere Mitglieder der PC-Familie teilen strukturelle Ähnlichkeiten, zu denen eine heterogene ∽10 kDa-Amino-Terminal-Proregion, eine hochgradig konservierte ∽55 kDa-Subtilisin-ähnliche katalytische Domäne und eine Carboxyl-terminale Domäne gehören, die in Länge und Reihenfolge heterogen ist. Diese Enzyme werden nach der Spaltung der Proregion im entsprechenden Zellraum katalytisch aktiv. Furin ist das einzige bekannte PC, das eine Transmembran-Domäne besitzt. Die Aufspaltung der Zielproteine erfolgt am Carboxyl-Terminus der Furain-Konsenssequenz RX(K/R)R. Es hat sich gezeigt, dass die saure Peptidsequenz C771 PSDSEEDEG780 Furin an das trans-Golgi-Netzwerk lokalisiert. Die Phosphorylation von Serinresten in dieser Region moduliert das intrazelluläre Routing von Furin-Proteinen. Eine weitere Signaldomäne ist die Tetrapeptid-Sequenz Y759KGL762, die die Internalisierung von der Zelloberfläche aus steuert.Spezifikation
