The Human MIP-3 beta/CCL19 ELISA quantitates Hu MIP-3β in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu MIP-3β. Principle of the method The Human MIP-3β solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. CCL19, auch MIP-3beta (Macrophage Inflamatory Protein3 beta) genannt, ist ein Mitglied der CC-Unterfamilie der Chemokine. Es ist am engsten mit CCL21 verwandt, mit dem es 32 % der Aminosäurensequenzhomologie teilt. CCL19 und CCL21 werden hauptsächlich durch stromale Zellen in den T-zellreichen Zonen von Lymphknoten exprimiert. Sie sind wichtige Vermittler des homeostatischen Transports von naiven T-Zellen und aktivierten dendritischen Zellen in die sekundären lymphatischen Organe. CCL19 und CCL21 spielen auch eine Rolle bei T-Zellen-Priming und -aktivierung sowie bei der Rekrutierung von Lymphozyten für entzündete Gewebe. Beide Proteine signalisieren über den G-Protein-gekoppelten Rezeptor CCR7, der auf T-Zellen und reifen dendritischen Zellen exprimiert wird. Obwohl CCL19 und CCL21 beide dieselbe Affinität für CCR7 aufweisen, führt nur die Bindung von CCL19 zur Desensibilisierung und Internalisierung des Rezeptors.Spezifikation
