The Human Serum Amyloid A (Hu SAA) ELISA quantitates Hu SAA in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu SAA. Principle of the method The Human SAA solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Serumamyloid A (SAA)-Proteine bestehen aus einer Familie kleiner (12 - 14 kDa, 104 - 112 Aminosäurereste), differentiell exprimierter Proteine, die bei Wirbeltieren stark konserviert werden. SAA-Proteine sind an den akuten Phasenreaktionen beteiligt; dies sind die unmittelbaren frühen Wirtsreaktionen auf Entzündungen. Während der akuten Phase werden die zirkulierenden SAA-Spiegel um das 100- bis 1000-fache erhöht und erreichen Konzentrationen von bis zu einem Milligramm pro Milliliter. Die Leber ist der Hauptort der SAA-Synthese, obwohl auch eine extrahepatische Expression berichtet wurde. Die Maus-SAA-Genfamilie ist auf Chromosom 7 abgebildet. In der Maus wurden fünf SAA-Gene und vier Proteinprodukte identifiziert: Maus-SAA1, -SAA2 und -SAA3 sind die akuten Phasenisoformen, SAA5 wird konstitutiv exprimiert, während SAA4 ein Pseudogen ist. SAAs gehören zu einer Kategorie von akuten Phasenproteinen, die auch C-reaktives Protein (CRP), Haptoglobin, Ceruloplasmin, die Komplementkomponenten C3 und C4, ein 1-Acid-Glykoprotein, einen 1-Proteinase-Inhibitor sowie Fibrinogen umfasst.
