MAP2K3 was assayed in a two stage coupled reaction involving phosphorylation of inactive MAPK14 (PV3305) by MAP2K3 followed by phosphorylation of MBP substrate by activated MAPK14. The enzymes were diluted in enzyme dilution buffer before adding to the reaction. In stage one, MAP2K3 (8.33μg/mL final concentration) is incubated with MAPK14 (66.7μg/mL final concentration) in 25mM Tris (pH 7.5), 10mM MgCl2, 0.5mM EGTA, 0.5mM Na3VO4, 5mM β-glycerophosphate, 2.5mM DTT, 0.01% Triton™ X-100, and 100μM ATP for 30 minutes at 30°C. In stage two, 5μL of the above reaction mix was transferred to 25μL of a reaction mixture containing 25mM Tris (pH 7.5), 10mM MgCl2, 0.5mM EGTA, 0.5mM Na3VO4, 5mM β-glycerophosphate, 2.5mM DTT, 0.01% Triton™ X-100, 100μM ATP, 20μg MBP substrate per reaction and trace [32P]-γ-ATP and incubated for 10 minutes at 30°C | |
