The Mouse CD30L (TNFSF8) ELISA quantitates Ms CD30L in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms CD30L. Principle of the method The Mouse CD30L solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Der Ligand für CD30 ist CD30L (CD153). Die Bindung von CD30 an CD30L vermittelt pleiotrope Effekte wie Zellvermehrung, -aktivierung, -differenzierung und apoptotischen Zelltod. CD30 spielt eine entscheidende Rolle in der Pathophysiologie der Hodgkins-Krankheit und anderer CD30+-Lymphome. CD30 fungiert als kostimulatorisches Molekül in der thymischen negativen Selektion. Neben seiner Expression auf Hodgkins- und Reed-Sternberg-Zellen findet sich CD30 auch in einigen Non-Hodgkins-Lymphomen (einschließlich Burkitts Lymphomen), virusinfizierten T- und B-Zellen sowie auf normalen T- und B-Zellen nach der Aktivierung. In T-Zellen ist die CD30-Expression auf den Teil der T-Zellen beschränkt, die Zytokine des Typs Th2 produzieren, und auf CD4+/CD8+-Thymozyten, die CD45RO und den IL4-Rezeptor coexprimieren.
