The Mouse E-Cadherin (CDH1) ELISA quantitates Ms E-Cadherin in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms E-Cadherin. Principle of the method The Mouse E-Cadherin solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. E-Cadherin (epitheliales Cadherin) ist ein klassisches Cadherin aus der Superfamilie der Cadherine (Calcium-abhängige Adhäsionsproteine). Das kodierte Protein ist ein Calcium-abhängiges Zelladhäsionsglykoprotein, das aus fünf extrazellulären Cadherin-Wiederholungen, einer Transmembranregion und einem stark konservierten zytoplasmatischen Schwanz besteht. Mutationen in diesem Gen korrelieren mit Magenkrebs, Brust-, Kolorektal-, Schilddrüse- und Ovarialkarzinom. Es wird angenommen, dass der Funktionsverlust zur Progression des Krebses beiträgt, indem die Proliferation, Invasion und/oder Metastasierung erhöht wird. Die Ektodomäne dieses Proteins vermittelt die bakterielle Adhäsion an Säugetierzellen und die zytoplasmatische Domäne ist für die Internalisierung erforderlich. Identifizierte Transkriptvarianten entstehen aus Mutationen an Consensus-Spleißstellen. E-Cadherin spielt eine zentrale Rolle beim Wachstum und der Entwicklung von Zellen, indem es die Gewebestruktur und die Erhaltung der Gewebeintegrität kontrolliert. Beim Menschen wird E-Cadherin durch das auf Chromosom 16 vorhandene CDH1-Gen kodiert. Studien haben gezeigt, dass die Reduktion und/oder der Verlust der E-Cadherin-Expression in Karzinomen positiv mit dem Potential zur Invasion und Metastasenbildung dieser Tumore zusammenhängt.Spezifikation
