The Mouse IGF Binding Protein 3 (Ms IGFBP-3) ELISA quantitates Ms IGFBP-3 in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms IGFBP-3. Principle of the method The Mouse IGFBP-3 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Insulin wie das Wachstumsfaktor-Bindungsprotein3 (IGFBP3) ist ein Mitglied der Superfamilie der insulinähnlichen Wachstumsfaktor-Bindungsproteine (IGFBP), die sechs hochaffine IGF-Bindungsproteine (IGFBP) und mindestens vier niederaffine Bindungsproteine (auch als IGFBP-bezogenen Proteine oder IGFBPrP bezeichnet) enthalten. Die IGFBP-Mitglieder sind Zystein-reiche Proteine mit konservierten Zystein-Rückständen, die im Amino-Terminal und in den Carboxy-terminalen Regionen des Moleküls gruppiert sind. Die cDNA-Sequenz, die das reife humane IGFBP3 kodiert, wird mit dem Signalpeptid von CD33 fusioniert. Der 265-Aminosäurerest rekombinanter reifer humaner IGFBP3 hat eine berechnete Molekularmasse von ungefähr 29 kDa. Aufgrund der Glykosylierung wandert das rekombinante Protein mit 41 kDa in SDS-PAGE.
