The Human FAK (Total) ELISA Kit is designed to detect and quantify the level of FAK (Total) in fresh or frozen human, mouse or rat cell lysates. The assay recognizes both natural and recombinant FAK (Total), independent of its phosphorylation state. Principle of the method A monoclonal capture antibody specific for FAK (Total) has been coated onto the wells of the 96-well plate. During the first incubation, standards of known content and unknown samples are pipetted into the wells and the antigen binds to the immobilized (capture) antibody. After washing, a rabbit antibody specific for the target protein is added to the wells and serves as a detection antibody by binding to the immobilized protein captured during the first incubation. After washing, a horseradish peroxidase labeled anti-rabbit IgG is added. This binds to the detection antibody to complete the four member sandwich. After a third incubation and washing to remove all the unbound enzyme, a substrate solution (TMB) is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of target protein present in the original specimen and the optical density can be read on a standard microplate reader. Fokale Adhäsionskinase (FAK) ist eine 125 kDa-Nicht-Rezeptor-Protein-Tyrosinkinase, die als Substrat für Src fungiert und ein wichtiges Element der Integrin-Signalübertragung ist. FAK spielt eine wichtige Rolle für Zellverteilung, -differenzierung, -migration, Zelltod sowie Beschleunigung des Phasenübergangs von der G1- zur S-Phase des Zellzyklus. Tyrosin 397 ist die Autophosphorylierungsstelle von FAK und an dessen anfänglicher Aktivierung beteiligt. Diese phosphorylierte Stelle bindet an die SH2-Domänen der Src-Familie und die p85-Untereinheit der PI3-Kinase und aktiviert die Zellmigration und -invasion. FAK hat eine zentrale katalytische Domäne und einen C-terminalen Schwanz, der es an den fokalen Adhäsionen lokalisiert. Dies sind Stellen, an denen Zellen über Oberflächen-Integrin-Rezeptoren an die extrazelluläre Matrix angedockt sind. Eine erhöhte FAK-Tyrosinphosphorylierung tritt auf, wenn Integrin mit Fibronektin eintritt. Die Adhäsion von Maus-NIH3T3-Fibroblasten an Fibronektin fördert die Assoziation des Grb2-Adapterproteins und c-Src-PTK mit FAK in vivo und führt auch zur Aktivierung der ERK2-MAP-Kinase. In v-Src-transformiertem NIH3T3 ist die Assoziation von v-Src, Grb2 und Sos mit FAK unabhängig von der Zelladhäsion an Fibronektin. In vitro bindet die Grb2-SH2-Domäne direkt an Tyrosin-phosphoryliertes FAK, die Bindungsstelle wurde durch Bindungsstellen-gerichtete Mutagenese als Tyr925 identifiziert. Für das FAK-Gen wurden mehrere Transkriptvarianten gefunden, die für unterschiedliche Isoformen codieren, jedoch wurden nur von dreien die Volllänge-Eigenschaften bestimmt.
