The Rat Epidermal Growth Factor (Rt EGF) ELISA quantitates Rt EGF in rat serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Rt EGF. Principle of the method The Rat EGF solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. EGF gehört zu einer Familie von Wachstumsfaktoren, die an den 170 kDa-EGF-Rezeptor gebunden sind. Die EGF-Rezeptorliganden werden als Typ I-Transmembranproteine mit 130 kDa mit einer N-terminal-Erweiterung, dem EGF-Modul, und einem C-terminal-Fragment, auch als zytoplasmatischer Schwanz bezeichnet, synthetisiert. EGF umfasst neun EGF-Motive; nur der neben der Zellmembran liegende Bereich ist als EGF-Rezeptor-bindende Domäne funktionsfähig. Aufgrund der proteolytischen Spaltung wird ein löslicher Wachstumsfaktor mit 6 kDa, der das EGF-Modul enthält, in den extrazellulären Raum freigesetzt (′Ektodomänenabwurf′). EGF kann in den meisten Körperflüssigkeiten nachgewiesen werden; die Konzentration ist in Plasma, Urin, Speichel und Milch ist besonders hoch. Thrombozyten sowie Zellen in verschiedenen Organen wie Gehirn, Niere, Speicheldrüse und Magen setzen EGF frei. Als Mitogen für endotheliale Zellen wirkt sich EGF auch auf die Angiogenese aus und spielt eine Rolle bei Wundheilungsprozessen. Die Blockierung der Freisetzung von EGF-Rezeptorliganden hemmt Wachstum und Migration in mehreren EGF-Rezeptor-abhängigen Zelllinien und verzögert die Wundreepithelialisierung aufgrund einer beeinträchtigten Keratinozytenmigration erheblich. Die Überexpression eines oder mehrerer Rezeptoren und/oder Liganden ist ein Merkmal der meisten Karzinome und Epithelkarzinome beim Menschen.Spezifikation
