Centrifuge 5425 +Rotor FA-18x2-Kit
Target antigen production, sequencing sample preparation, and antibody humanization involve a great deal of repetitive molecular biology handling steps. This includes spin column based MiniPrep kits for nucleic acid purification, gel extraction, and PCR clean-up. Each protocol has multiple centrifugation steps interrupted by pipetting and vessel changes. Microcentrifuges have entered a new generation, where the completely newly developed Centrifuge 5425 in combination with kit rotor specifically designed for 1.5/2.0 mL spin column kits.
Centrifuge 5910 R + Rotor FA-6x50
Lysate clarification in nucleic acid purification protocols for target antigen production and antibody humanization include high speed centrifugation runs with up to 20,000 x g. High numbers of temperature-sensitive samples with up to 50 mL need to be processed. Each protocol has multiple centrifugation steps interrupted by pipetting and vessel changes. The production of antibodies in cell lines and antigens in bacteria involve harvesting cells from large volume suspensions of several liters. Purifying peripheral blood mononuclear cells (PBMC), FACS/Flow cytometry and hybridoma technology include multiple centrifugation steps of temperature-sensitive samples in many different consumables. This requires great versatility and an advanced temperature management. In addition, PBMC isolation by Ficoll gradient centrifugation must be carried out with a minimum of vibration during deceleration (breaking) within reasonable time. For faster high-yield PBMC isolation, acceleration and deceleration ramps can be selected individually.

High-throughput screening for the identification of antibody hits in culture supernatants of hybridoma clones is usually performed in microtiter plates (MTPs) with 96 or more wells. Various methods are required and all include multiple centrifugation steps of temperature-sensitive samples in MTPs. Refrigerated benchtop Centrifuge 5920 R in combination with swing-bucket Rotor S-4x1000 with plate/tube buckets offer super-high capacity of up to 28 MTPs per run and an advanced cooling performance for optimal sample protection.
Centrifuge 5430 + Rotor A-2-MTP
For the selection and characterization of antibody leads low-throughput screening assays are performed in microtiter plates (MTPs). Various methods are required and all include multiple centrifugation steps. Centrifuge 5430 in combination with swing-bucket Rotor A-2-MTP offer an economical solution for two MTPs per run in a small footprint. Centrifuge 5430 also accommodate rotors for 15 mL and 50 mL conical tubes and high-capacity rotors for 1.5/2.0 mL tubes.
Centrifuge 5810 + Rotor S-4-104 + 4 x 5/15 mL adapter + 4 x 50 mL adapter
In a final step of the mAb drug discovery workflow a stable cell line is established that can express high amounts of humanized monoclonal antibody long-term. Cultivation and selection of stable cell lines require multiple centrifugation steps. Centrifuge 5810 in combination with Rotor S-4-104 offer an economical solution with an excellent footprint to capacity ratio. This setup is ideal for standard cell culture applications including cell harvesting in 15 mL or 50 mL conical tubes.
