Fixation: Samples can be stored in IC Fixation Buffer (Product No. 00-8222-49) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product No. 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. It is abundantly expressed on the surface of nearly all human peripheral blood lymphocytes. The CD52 antigen is GPI-anchored membrane receptor that consists of a small 12 amino acid peptide attached to a large N-linked carbohydrate group. CD52 is a potent target for inducing cytolysis of mature lymphocytes. Alemtuzumab, a humanized monoclonal antibody targeting CD52, is used as an immunomodulatory drug for the treatment of chronic lymphocytic leukemia, T cell lymphomas, multiple sclerosis, and other diseases.
