Description

Effektives Screening von Kolonien mit X-Gluc, einem Substrat für ß-Glucuronidase (GUS), die von gusA, einem gängigen Reportergen, kodiert wird.

X-Gluc (5-brom-4-chlor-3-indolyl-beta-D-glucuronsäure, Cyclohexylammonium-Salz) ist ein Substrat für Beta-Glucuronidase (GUS), das von gusA, einem verbreitet eingesetztem Reportergen kodiert wird. Glucuronidase spaltet X-Gluc und bildet so die farblose Gluconsäure und ein intensiv blaues Chlor-Brom-Indigo-Präzipitat (siehe nachstehende Abbildung).

Empfohlen für:

  • Nachweis der GUS-Expression in Pflanzenzellen und -geweben (1)
  • Nachweis von Infektionen durch E. coli
  • Nachweis bakterieller Kontaminationen in Lebensmittel- und Wasserproben

Hinweis:

Zubereitung einer 0,1 M Stock-Lösung in Dimethylsulfoxid wird empfohlen.
