Beschreibung

Mevastatin (Compactin) inhibits HMGCR (HMG-CoA reductase) , induces apoptosis which in turn inhibits isoprenoid biosynthesis and therefore blocks protein isoprenylation and reduces plasma cholesterol levels in humans. Mevastatin causes cells to arrest early in the G1 phase. Mevastatin is a close structural analog of lovastatin and both agents have the same biochemical and pharmacological activities. Mevastatin is inactive in cell-free assays. In cells however, it is hydrolyzed to the active free acid form by intracellular esterases.

Anwendungen
Mevastatin (Compactin) hemmt HMGCR (HMG-CoA-Reduktase) und induziert Apoptose, die wiederum die Isoprenoid-Biosynthese hemmt, daher die Proteinisoprenylierung blockiert und den Plasmacholesterinspiegel beim Menschen senkt. Mevastatin bewirkt, dass Zellen früh in der G1-Phase verbleiben. Mevastatin ist ein enges strukturelles Analogon von Lovastatin. Beide Wirkstoffe haben die gleichen biochemischen und pharmakologischen Eigenschaften. Mevastatin ist in zellfreien Assays inaktiv. In Zellen wird es jedoch durch intrazelluläre Esterasen zur aktiven freien Säureform hydrolysiert.

Löslichkeit
Löslich in Ethanol bei 25 mg/ml und in DMSO bei 20 mg/ml
