Kit is designed to detect and quantify the level of STAT3 (Phospho) [pY705] in fresh or frozen human cell lysates. Cross-reactivity has been observed in mouse and rat cells. The assay recognizes both natural and recomb. human STAT3 (Phospho) [pY705]. Principle of the method A monoclonal capture antibody specific for STAT3 (Phospho) [pY705]) has been coated onto the wells of 96-well plate. During the first incubation, standards of known content and unknown samples are pipetted into the wells and the antigen binds to the immobilized (capture) antibody. After washing, a rabbit antibody specific for the target protein is added to the wells and serves as a detection antibody by binding to the immobilized protein captured during the first incubation. After washing, a HRP labeled anti-rabbit IgG is added. This binds to the detection antibody to complete the four member sandwich. After a third incubation and washing to remove all the unbound enzyme, a substrate solution (TMB) is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of target protein present in the original specimen and the optical density can be read on a standard microplate reader. STAT3 gehört zur Familie der STAT-Proteine (Signalwandler und Transkriptionsaktivatoren), die durch rezeptorassoziierte Kinasen phosphoryliert, zum Zellkern transloziert und als Transkriptionsfaktoren bei der Reaktion auf Zytokine und Wachstumsfaktoren fungieren. Koaktivatoren wie CREB-bindendes Protein sind für die transkriptionelle Aktivierung durch STAT3 erforderlich. STAT3 kann auch durch Interferon-alpha, Interferon-gamma, EGF, PDGF und IL6 aktiviert werden. Die Phosphorylierung auf Tyrosin705 durch JAK1 und JAK2 ist für die STAT3-Dimer-Bildung, die nukleare Translokation und die DNA-Bindungsaktivität unerlässlich. Beim Menschen befindet sich das STAT3-Gen auf dem q-Arm des Chromosoms 17. STAT3 wird nachweislich durch IFN-alpha aktiviert, nicht jedoch durch IFN-beta. Die mit STAT3 assoziierten Transkriptionsfaktoren sind c-Jun und zyklisches AMP-reaktives Bindungsverstärkerprotein (CREB). STAT3 vermittelt die Expression einer Vielzahl von Genen als Reaktion auf Zellreize und spielt somit eine Schlüsselrolle bei vielen zellulären Prozessen wie Zellwachstum und Apoptose. Bei der kleinen GTPase Rac1 hat sich gezeigt, dass sie die Aktivität von STAT3 bindet und reguliert, während das PIAS3-Protein ein spezifischer Inhibitor von STAT3 ist. Drei alternativ gespleißte Transkriptvarianten, die verschiedene Isoformen von STAT kodieren,3 wurden beschrieben. Die Löschung des STAT3-Gens bei Knockout-Mäusen ist im frühen embryonalen Stadium tödlich.
