DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12. - DMEM (Dulbecco's Modified Eagle Medium) ist ein weit verbreitetes Basalmedium zur Wachstumsunterstützung vieler verschiedener Säugetierzellen.
- Zu den erfolgreich in DMEM kultivierten Zellen gehören primäre Fibroblasten, Neuronen, Gliazellen, Endothelzellen aus humanen Nabelschnurvenen (HUVEC) und glatte Muskelzellen sowie Zelllinien wie HeLa, 293, COS-7 und PC-12.

- Ohne: Natriumpyruvat, HEPES
Verwendung von DMEM
- DMEM enthält eine viermal höhere Konzentration an Aminosäuren und Vitaminen als das ursprüngliche Eagle's Minimal Essential Medium.
- Es wurde ursprünglich in einer Formulierung mit niedriger Glukosekonzentration (1 g/l) und mit Natriumpyruvat entwickelt, wird aber häufig mit höheren Glukosekonzentrationen sowie mit oder ohne Natriumpyruvat eingesetzt.
- DMEM enthält keine Proteine, Lipide oder Wachstumsfaktoren. Daher ist eine Supplementierung erforderlich, üblicherweise mit 10 % fötalem Kälberserum (FKS).
- DMEM verwendet ein Puffersystem mit Natriumbikarbonat (3,7 g/l) und erfordert daher eine 5 bis 10%ige CO2-Umgebung, um den physiologischen pH-Wert aufrechtzuerhalten.
