ADAMTS (a disintegrin and metalloproteinase with thrombospondin motif) is a family of secreted proteinases related to, but distinct from ADAM (a disintegrin and metalloproteinase). Presently eighteen family members have been identified in the human genome. ADAMTS1 consists of a prodomain, a metalloprotease domain, a thrombospondin (TSP) homologous domain with TSP type I motif, a spacer region and C-terminal TSP submotifs. The prodomain is cleaved off by a furin-type protease. There occurs additional processing at the C-terminal end. ADAMTS1 has been found in human articular cartilage and in neurons. The protein is induced by IL-1. ADAMTS1 displays antiangiogenic properties and inhibits endothelial cell proliferation. ADAMTS1-null mice exhibit growth retardation and defects in morphology. The active protease cleaves aggrecan at several sites and versican. ADAMTS1 activity is inhibited by metalloproteinase inhibitors TIMP 3 and TIMP 4 at higher concentrations and by catechin gallate esters. RP-77543 is a purified, truncated recombinant human ADAMTS1 protein (amino acids F253-N616 with a C-terminal His tag), expressed in insect cells. Purity is greater than 90% by SDS-PAGE. Calculated Molecular weight is 41 kDa, actual SDS-PAGE results show a band at ∽50 kDa. Specific Activity: Activity of ADAMTS1 is determined with recombinant aggrecan interglobular domain (Product &Num; RP-77527). ADAMTS1 hydrolyzes the "aggrecanase" site within this domain (peptide bond E373 -A374 in human aggrecan). The hydrolysis rate is greater than 0.06 nM hydrolyzed substrate/min x nM truncated ADAMTS1. Permg of enzyme, activity is greater than 1.4 nmoles hydrolyzed substrate/min xmg ADAMTS1. The recombinant substrate is incubated at a concentration of 0.1μM with 0.2-3.0 nM ADAMTS1 in 50 mM Tris-HCL, pH 7.5, 150mM NaCl, 5mM CaCl2, 1 μM leupeptin, 1 μM pepstatin, 1 mM Pefabloc, and 0.05% Brij 35 for 15 min at 37°C. Cleavage is estimated from the appearance of the hydrolysis fragment with N-terminus ARGSVIL. RP-77543 may be used as a control with corresponding anti-ADAMTS1 antibodies. ADAMTS1 is inhibited by chelators of divalent cations such as EDTA and catechin gallate esters.Spezifikation
