The Human Macrophage-Colony Stimulating Factor (CSF-1) (Hu M-CSF) ELISA quantitates Hu M-CSF in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu M-CSF. Principle of the method The Human M-CSF solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. M-CSF (Makrophage-Kolonie-stimulierender Faktor, CSF-1) ist ein Überlebensfaktor, der für die Proliferation und Entwicklung von Monozyten, Makrophagen und Osteoklast-Vorläuferzellen unerlässlich ist. M-CSF induziert auch VEGF (vaskulärer endothelialer Wachstumsfaktor)-Sekretion durch Makrophagen und vermittelt dadurch die Mobilisation von Endothel-Vorläuferzellen und Neovaskularisation. M-CSF gibt es in mehreren bioaktiven Isoformen, die sich in ihrer Wirksamkeit und Stabilität unterscheiden. Das Protein wird in ganzer Länge als membranübergreifendes Protein synthetisiert, das auf der Zelloberfläche exprimiert oder im sekretorischen Vesikel weiter gespalten und modifiziert werden kann. Darüber hinaus ist M-CSF ein disulfid-gebundener Homodimer, der zu einer von zwei Isoformen, einem Glykoprotein oder einem Proteoglycan, verarbeitet wird, das durch Zugabe von Chondroitinsulfat zu jeder Untereinheit modifiziert wurde. Die Bindung von M-CSF an seinen Rezeptor, c-Fms (CSF-1R oder CD115) induziert die Dimerisierung des Rezeptors, gefolgt von der Internalisierung und dem Abbau des Komplexes. Funktionell ist M-CSF dafür bekannt, dass es die Differenzierung von hämatopoetischen Stammzellen zu monozyten Makrophagen-Zellpopulationen in der Kultur stimuliert. M-CSF wirkt durch den CSF-Rezeptor1. Obwohl die menschliche M-CSF Aktivität auf Mauszellen zeigt, zeigt die Maus-CSF keine Aktivität auf menschlichen Zellen.Spezifikation
