The Human Pepsinogen II (Hu PGC) ELISA quantitates Hu PGC in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu PGC. Principle of the method The Human PGC solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Pepsin ist eines der wichtigsten proteolytischen Enzyme, die von der Magenschleimhaut abgesondert werden. Pepsin besteht aus einer einzelnen Polypeptidkette und entsteht aus seinem Vorläufer, Pepsinogen, durch Entfernung eines 41-Aminosäuresegments aus dem N-Terminus. Pepsinogen wird im Magenfutter synthetisiert. Von der Magenschleimhaut ebenfalls produzierte Flusssäure ist notwendig, um das inaktive Enzym umzuwandeln und die optimale Säure (pH1 -3 ) für die Pepsin-Funktion zu erhalten. Pepsin ist besonders wirksam bei der Spaltung von Peptidbindungen mit aromatischen Aminosäuren. Pepsin weist eine extrem breite Spezifität auf, und obwohl Bindungen mit Phenylalanin und Leucin bevorzugt werden, werden viele andere auch in gewissem Maße gespalten. Die Aminosäurezusammensetzung von Pepsin C unterscheidet sich von der von Pepsinogen und Pepsin insbesondere im Gehalt an basischen Aminosäuren, Glutaminsäure, Asparaginsäure, Leucin und Isoleucin.Spezifikation
