The Human Neuronal Cell Adhesion Molecule-1(Hu N-CAM1) ELISA quantitates Hu N-CAM1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu N-CAM1. Principle of the method The Human N-CAM1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. CD56 (NCAM, neuronales Zelladhäsionsmolekül) ist ein transmembranes Glykoprotein der Immunglobulin-Familie, das als adhäsives Molekül dient und im Nervensystem in Isoformen von 120 bis 180 kDa ubiquitär exprimiert wird. CD56 wird auf T-Zellen und NK-Zellen gefunden und ist an Zellmigration, axonalem Wachstum, Wegfindung und synaptischer Plastizität beteiligt. Die Modifikation der Polysialinsäure führt zu einer Reduktion der CD56-vermittelten Zelladhäsion. Durch seine extrazelluläre Region vermittelt CD56 homophile und heterophile Wechselwirkungen durch Bindung extrazellulärer Matrixkomponenten wie Laminin und Integrine. CD56 wird auf den meisten neuroektodermalen Zelllinien, Geweben und Neoplasmen wie Retinoblastom, Medulloblastom, Astrozytomen und Neuroblastom exprimiert. Außerdem ist CD56 ein weit verbreiteter neuroendokriner Marker mit hoher Empfindlichkeit für neuroendokrine Tumore und ovarielle Granulosazelltumore. Zu den mit CD56-Dysfunktion assoziierten Krankheiten gehören Tollwut und blastische Plasmazytoide dendritische Zellneoplasmen.
