Kit is designed to detect and quantify the level of STAT5 alpha (Total) in fresh or frozen human cell lysates. Cross-reactivity has been observed in mouse and rat cells. The assay recognizes both natural and recomb. human STAT5 alpha (Total), independent of its phosphorylation state. Principle of the method A monoclonal capture antibody specific for human STAT5 alpha (Total) has been coated onto wells of 96-well plate. During the first incubation, standards of known content and unknown samples are pipetted into the wells and the antigen binds to the immobilized (capture) antibody. After washing, a rabbit antibody specific for the target protein is added to the wells and serves as a detection antibody by binding to the immobilized protein captured during the first incubation. After washing, a HRP labeled anti-rabbit IgG is added. This binds to the detection antibody to complete the four member sandwich. After a third incubation and washing to remove all the unbound enzyme, a substrate solution (TMB) is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of target protein present in the original specimen and the O.D. can be read on a standard microplate reader. STAT5 Alpha ist ein Mitglied der STAT Familie der Transkriptionsfaktoren. Als Reaktion auf Zytokine und Wachstumsfaktoren werden die Mitglieder der STAT-Familie durch die mit dem Rezeptor verbundenen Kinasen phosphoryliert und bilden dann Homo- oder Heterodimere, die in den Zellkern transplantiert werden, wo sie als Transkriptionsaktivatoren wirken. STAT5 alpha wird aktiviert durch und vermittelt die Reaktionen vieler Zellliganden, wie IL2, IL3, IL7 GM-CSF, Erythropoietin, Thrombopoietin und verschiedener Wachstumshormone. Die Aktivierung von STAT5 alpha bei Myelom und Lymphom im Zusammenhang mit einer TEL/JAK2-Genfusion ist unabhängig von Zellreiz und hat sich als wesentlich für die Tumorgenese erwiesen. Das Maus-Gegenstück von STAT5 alpha wird gefunden, um die Expression von BCL2 L1/BCL-X(L) zu induzieren, was die antiapoptotische Funktion dieses Proteins in Zellen suggeriert. STAT5 alpha, zusammen mit STAT5 beta, wurden in der Maus identifiziert. Die Aminosäurensequenzen von STAT5 alpha und STAT5 beta weisen eine 96 %-Sequenzähnlichkeit auf, und beide Proteine werden in den meisten Geweben von jungfräulichen und laktierenden Mäusen koexprimiert.
