The Monkey PAI-1 (SERPINE1) ELISA quantitates Rhesus Mk PAI-1 in monkey serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Mk PAI-1. Principle of the method The Monkey PAI-1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. PAI1 (Plasminogen-Aktivator-Inhibitor1) gehört zur Superfamilie der Serinprotease-Inhibitoren und ist der Hauptinhibitor von Gewebetypen und Urokinase-Plasminogenaktivatoren (tPA und uPA). Thrombozyten sind die Hauptquelle der zirkulierenden PAI11, werden aber von vielen Gewebe- und Zelltypen synthetisiert und sezerniert, darunter Fibroblasten, glatte Muskelzellen, Endothelzellen, Hepatozyten und entzündliche Zellen. Die Expression von PAI11 kann auf transkriptioneller Ebene durch viele Faktoren reguliert werden, darunter Wachstumsfaktoren, Zytokine, Hormone, entzündliche Faktoren, Glukose- oder Lipidmetaboliten, vaskuläre tonregulierende Faktoren, Chemikalien und andere Umwelt- oder physikalische Faktoren. PAI1 ist in verschiedenen Krankheitszuständen mit erhöhten Konzentrationen vorhanden und wurde mit einem erhöhten Auftreten von Thrombosen bei Adipositas, Thrombophilie und dem metabolischen Syndrom in Verbindung gebracht. Defekte mit PAI-1 sind durch abnormale Blutungen gekennzeichnet. PAI1 vermittelt die Hemmung der Fibrinolyse durch Hemmung der Aktivität des Plasminogenaktivators und kann das neuronale Überleben fördern. Andere Defekte in PAI1 sind die Ursache für Plasminogenaktivator-Inhibitor1mangel (PAI-1Mangel).
