The Human Osteopontin/SSP1 ELISA quantitates Hu SSP1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu SSP1. Principle of the method The Human SSP1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Osteopontin ist ein Arginin-Glycin-Asparaginsäure (RGD)-haltiges Glykoprotein, das mit Integrinen und CD44 als Hauptrezeptoren interagiert. Es handelt sich um ein multifunktionelles Protein, das an der Knochenmineralisierung, Zelladhäsion, Zellmigration, chronischen entzündlichen Erkrankungen und der Transformation beteiligt ist. Die proteolytische Aufspaltung durch Thrombin- und Matrixmetalloproteinasen nahe der integratin-bindenden Arg-Gly-Asp-Sequenz moduliert die Funktion des OPN und seine integrin-bindenden Eigenschaften. Thrombin-gespaltene Fragmente von Osteopontin werden in bösartigen Glialtumoren überexprimiert, bieten eine molekulare Nische mit Überlebensvorteil und stellen ein neuartiges Substrat für Plasmin und Cathepsin D dar.Spezifikation
